By integrating molecular and clinical data, large-scale biobanks have become powerful resources for precision health. Their extensive sample sizes and rich phenotypic information enable the discovery of novel gene–disease associations, improved disease risk prediction, and the identification of causal biomarkers and therapeutic targets. In this talk, I will focus on genome-based risk prediction, with an emphasis on polygenic risk scores (PRS). I will first introduce the fundamental concepts of PRS and highlight recent methodological advances. In particular, I will discuss approaches that incorporate functional annotations, extend PRS to rare variants, and improve model interpretability. Beyond genome-based prediction, I will also present emerging strategies that leverage proteomic data for disease risk prediction and biological insight.
